Purification and Quality Control

The His-tag recombinant protein is purified by affinity chromatography in combination with FPLC columns. The purified CTF1 is greater than 90% homogeneous based on SDS-PAGE analysis.

Unit Definition (Activity)

1 unit equals 1 nanogram of purified protein. 30-100 units (ng) are required for a reconstituted transcription assay and 5-20 units (ng) are required for a gel mobility shift assay in a 20µl reaction.

Applications

Purified CTF1 has been used for in vitro transcriptional activation, DNase I protection, gel mobility shift and protein-protein interaction assays. For Research Use Only.

Formulation and Storage

The protein is in 20mM Tris-HCl pH7.9,100mM NaCl, 0.2mM EDTA, 1mM DTT and 20% glycerol. Stored at -70°C before use. Avoid repeated freeze thaw cycles.

Background

The CCAAT-box binding protein 1 (CTF1, also called NF-I or NF-1) is a proline-rich transcription activator (1,2). It selectively activates transcription of genes that contain GCCAAT consensus sequence in promoter region, including human Ha-Ras, alpha-globin, hsp70 and many other cellular and viral genes (1, 3-5). Cloning and functional characterization of the protein revealed that CTF1 contains an N-terminal DNA binding domain and a C-terminal proline-rich activation domain (2,3). CTF1 was shown to interact with the general transcription factor IIB (TFIIB) and facilitate the recruitment of TFIIB to TBP-DNA complexes (6), with the forkhead thyroid transcription factor TTF2 to regulate hormone–dependent transcription of thyroperoxidase gene (7), as well as other receptor proteins to synergize hormone receptor-responsive activation (8-10).
