Purification and Quality Control
The His-tag recombinant protein is purified by affinity chromatography in combination with FPLC columns.
The purified ERK2 is greater than 95% homogeneous based on SDS-PAGE analysis.

Applications
Recombinant ERK2 can be used for in vitro phosphorylation, dephosphorylation assays and inhibitor screening assay.
Formulation and Storage
The protein is in 20mM Tris-HCl pH7.9,100mM NaCl, 0.2mM EDTA, 1mM DTT and 20% glycerol. Stored at -70°C before use. Avoid repeated freeze thaw cycles.

Background
The extracellular signal-regulated kinase 2 (ERK2, 42kDa), also known as mitogen-activated protein kinase 1 (MAPK1), is located in an integration point of the mitogen-activated protein kinase (MAPK) cascading pathways and have been implicated in the regulation of diverse cellular processes such as proliferation, differentiation, transcription regulation and development (1-4). Activation and inactivation of ERK2, a serine/threonine protein kinase, are tightly regulated by an upstream protein kinase MEK1/2 and dual-specificity protein phosphatases (DSUP). In human cancers and human disorders, aberrantly upregulated MEK1 activates its unique substrate ERK1/2. Therefore, the Ras-Raf-MEK1-ERK2 pathway has become an attractive target for anticancer therapeutics (5).
