High efficiency recombinant ProTie Liagse was used as a molecular "stapler" to link proteins with the C-terminal LPETGXn motif to other proteins or molecules possessing a GGGG motif at their N terminal.
1 kit have ligase and reaction buffer that is enough for 2mg target protein ligation.
High efficiency ProTie ligase can work at 4°C and efficiency could reach up to 90%.
ProTie ligase is a bacterial transpeptidase that covalently attaches proteins to the bacterial cell wall by cleaving between threonine and glycine at an LPXTG recognition motif and catalyzes the formation of an amide bond between the carboxyl-group of threonine and the amino-group of the cell-wall peptidoglycan.
Recombinant ProTie ligase is highly optimized Sortase A from Staphylcoccus aureus. It was expressed in E. coli expression system with a N-terminal His tag. Its MW is 21.7-kDa. The purity is >95% as estimated on SDS PAGE.
One ug of recombinant ProTie ligase can ligate 100ug target proteins.
Not only this kit can be used for protein protein liagtion, it also can be used for fusion protein removal, N-terminal labeling, peptide cyclization, DNA-protein hybrid, etc.
